Progress Report on LC - What's the hold up? - Mushroom Cultivation (2024)

So after getting my flow cube, I've had zero contam issues which has been GREAT. My first go around was inoculating brown rice jars with Golden Teacher liquid spores and they all went bad, super gross. Retired the SAB and got a flow cube. All my agar plates have been perfect since then.

I took a clean sample of Nepal and dropped it into LC made with malt. It has flourished.

Took a bit of that LC and put into a WBS jar and this is after 6 days. Looking right?

Company I got the spore syringes from sent me a replacement for the GT one, and I put that straight into the LC jars I made with the same recipe and they both look wonderful, imo. Did those on 3/25

Did a quality check for contams on agar and this is what it's lookin like now.

So my confusion/concern is on the Koh Samui I did the exact same way with liquid spores into liquid culture. I split one syringe into two jars made the same way on 3/29 and I haven't seen any growth at all. They look just a clear as they did on day 1.

Thoughts? I would think I'd see some action by now considering the progress on the GT ones.

By “flow cube”, I presume you mean flowhood.

The procedures you followed with the Nepal is the proper way to inoculate LC. Although the LC you inoculated with the GC spores appears clean, I would not take it to grains until taking two more transfers from that GT plate first.

A liquid culture can appear clean but be contaminated. If your spore to LC ends up being clean, you got lucky. Spores to LC can work, but it is a gamble and should not be counted as proper procedure.

As far as the Koh Samui the spores could be old and no longer viable.

Sorry, "FloCube" is the brand of flow hood, but I spelled it wrong.

Just out of curiosity, do you have bad experiences with "guaranteed contaminant free" liquid spore syringes contaminating LC? It feels weird taking 10cc of liquid spores and using them directly on agar because of how much liquid ends up on the plate. I think I had like 15 plates last time I tried that with the Nepal.

Let's say I end up with two more transfers of clean GT, should I take another clean sample and start a new LC with it and toss the others? My main goal was to have a steady supply of mycelium without crowding my fridge with plates.
